Quantitative Determination of Twelve Bioactive Components in Mahwang-tang, a Traditional herbal Prescription, using an LC-MS/MS

Abstract
Mahwang-tang is a traditional herbal formula consisting of six herbal medicines has been used for the treatment of influenza. In this study, quantitative analysis of the twelve bioactive components, ephedrine(1), amygdalin(2), liquiritin apioside(3), liquiritin(4), ferulic acid(5), liquiritigenin(6), coumarin(7), cinnamic acid(8), kaempferol(9), cinnamaldehyde(10), glycyrrhizin( 11), and 6-gingerol(12) in Mahwang-tang decoction was performed using an liquid chromatography-electrospray ionization- mass spectrometer. The compounds 1?12 was separated using an UPLC BEH C18(100?2.1 mm, 1.7 ƒÊm) column and eluted with a gradient of 0.1%(v/v) aqueous formic acid(A) and acetonitrile(B) at 0.3 mL/min. The column oven temperature was set at 45oC and injection volume was 2.0 ƒÊL. Correlation coefficient of the compounds 1?12 ranged between 0.9949 and 0.9999. The limit of detection and quantification of the compounds 1?12 were 0.02?4.73 ng/mL and 0.06?14.20 ng/mL, respectively. The concentration of the compounds 1?12 in lyophilized decoction extract of Mahwang-tang were in the range 0.29 to 304.88 mg/g.
